چکیده

Optical measurements of the pigments of the retinal pigment epithelium (RPE) and choroid were made on 38 human autopsy eyes of both blacks and whites, varying in age between 2 wk and 90 yr old. Lipofuscin in melanin-bleached RPE was measured as fluorescence at 470 mm following excitation at 365 nm and was found to be proportional to fluorescence measured at 560 nm in unbleached tissue. Transmission measurements of RPE and choroidal melanin were converted and expressed as optical density units. The choroidal melanin content increased from the periphery to the posterior pole. RPE melanin concentration decreased from the periphery to the posterior pole with an increase in the macula. Conversely, the amount of RPE lipofuscin increased from the periphery to the posterior pole with a consistent dip at the fovea. There was an inverse relationship between RPE lipofuscin concentration and RPE melanin concentration. The RPE melanin content was similar between whites and blacks. Lipofuscin concentration was significantly greater (P = 0.002) in the RPE of whites compared to blacks; whereas blacks had a significantly greater (P = 0.005) choroidal melanin content than whites. The amounts of both choroidal and RPE melanin showed a trend of decreasing content with aging, whereas the amount RPE lipofuscin tended to increase (whites greater than blacks). Per fundus area, the amount of choroidal melanin was always greater than that in the RPE. There was a statistically significant (P = 0.001) increase in RPE height with age, most marked in eyes of whites after age 50 and correlated with the increase in lipofuscin concentration.

Lipofuscin and melanin of human retinal pigment epithelium. Fluorescence, enzyme cytochemical, and ultrastructural studies.

The life history of melanin and lipofuscin granules of human retinal pigment epithelium (RPE) was studied in 30 human eyes spanning nine decades of life. Autofluorescent granules in the cytoplasm of eye over 30 years of age were shown, ultrastructurally and through lipid solvent extraction, to be lipofuscin granules. Sparse small fluorescent granules in infant eyes were secondary lysosomes cont...

Aging human RPE: morphometric analysis of macular, equatorial, and peripheral cells.

Retinal pigment epithelium (RPE) of 50 human eyes, five from each 10 decades of life, were analyzed using ultrastructural morphometric techniques. Content of three types of pigments, lipofuscin, melanin, and complex granules, (melanolipofuscin, melanolysosomes) were recorded for cells from macular, equatorial, and peripheral retinal specimens. Areas occupied by pigments, nucleus, and cytoplasmi...

Tissue culture of human retinal pigment epithelium.

Adult human retinal pigment epithelium has been enzymatically isolated and established In tissue culture. Primary cultures survived three to six months. Occasionally, cell lines spontaneously evolved from primary cultures. Melanin granules decreased with the age of the culture, became less pigmented but retained some characterstics of melanin-producing cells. Lipofuscin granules are present, bu...

Oxidation causes melanin fluorescence.

PURPOSE The goal of this study is the characterization of the strong yellow fluorescence of oxidized melanin in the retinal pigment epithelium (RPE) and the choroid. METHODS Naturally occurring melanin in the human retina and choroid was oxidized by exposing fixed and plastic-embedded sections of a human eye to light and hydrogen peroxide. Synthetic melanin was also oxidized in vitro by expos...

Melanin concentration in normal human retinal pigment epithelium. Regional variation and age-related reduction.

Melanin concentrations were analyzed in the pigment epithelium of 61 postmortem normal human eyes from donors 14-97 yr of age. The content of soluble melanin in the pigment epithelium declined with age from the highest values of 95 micrograms/mg in the 14-50 yr age group to the lowest values of 22 micrograms/mg dry weight in the over 70 yr age group. In 16 of these eyes, regional measurements r...
